Neuroglial cells have several essential roles in the nervous sys-tem. Oligodendrocytes myelinate neurons accelerating impulsepropagation and supporting neuronal survival. Astrocytes con-tribute to formation of a privileged environment within the cen-tral nervous system (CNS). Dysfunctional neuroglia is observedin several neurodegenerative diseases. For example, demyelina-tion due to immune attack is the culprit of multiple sclerosis(MS). Response of astrocytes to immunoglobulin Gs (IgGs) iso-lated from amyotrophic lateral sclerosis (ALS) patients differfrom their response to IgGs isolated from controls. Our goal isto develop computer software to assist researchers in their analy-sis of myelin and astrocytes. First, existing and custom machinelearning based algorithms were compared in classification ofmyelin from fluorescent microscopy images. Both our customconvoluted neural network and support vector machine algo-rithms performed more accurately then other algorithms (over98% accurate). The methodology will be used for drug screensagainst demyelinating diseases such as MS. Second, we are eval-uating different segmentation strategies to accurately classifyastrocytes in time series images. We aim to identify astrocytesand extract calcium (Ca2+) dye intensity variation data over time.IgG from sera of ALS patients induce Ca2+ transients in culturedrat astrocytes. We aim to automate analysis of analysis of Ca2+intensity variation in order to develop IgG from sera of ALSpatient as diagnostic marker of the disease.
